消症汤对子宫内膜异位模型大鼠异位组织细胞增殖和凋亡的影响

EFFECTS OF XIAO ZHENG TANG ON PROLIFERATION AND APOPTOSIS OF ENDOMETRIC CELLS IN EMT MODEL RATS

目的 探讨“消症汤”对子宫内膜异位症 (endometriosis,EMT)模型大鼠异位组织细胞增殖和凋亡的影响 ,为临床用药提供实验依据。方法 制作大鼠EMT动物模型 ,模型鼠每日分别以 0 5ml和 1 0ml中药灌胃 ,连续用药 3周后观察异位组织细胞增殖指数和凋亡指数。结果 模型对照组细胞增殖指数大于用药组 ,凋亡指数小于用药组 (P <0 0 1)。 0 5ml组和 1 0ml组比较细胞增殖指数和凋亡指数无显著差异 (P >0 0 5 )。结论 “消症汤”能促进异位组织细胞凋亡 ,抑制异位组织增殖。

Objective To study the effects of a preparation of traditional Chinese medicine (Xiaozhengtang) on the proliferation and apoptosis of endometric cells in endometriosis (EMT) model rats. Method First, rat endometriosis model was produced. Second, three weeks after the operation, the EMT model rats were randomly divided into 3 groups: 2 experimental groups and 1 control group. The experimental groups were poured the Chinese medicine down the throat, 0.5ml and 1.0ml respectively every day for each rat. The control group was treated with 1.0ml H_ 2 O for each rat. Third, the changes of the normal and transplanted endometrium were observed. The proliferating cells and apoptotic cells in the endometrium were studied. The marker for proliferation is proliferating cell nuclear antigen (PCNA) immunohistochemical staining, while the apoptotic cells were detected by using DNA in situ terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick ending labeling (TUNEL). Result The proliferation index of endometriotic cells was 56.28%±1.90% in the control group, 21.04%±0.78% in the 0.5ml experimental group and 21.26%±0.83 %in the 1.0ml group. The apoptotic index was 20.08 %±0.38%in the 0.5ml group,and 20.74%±0.52%in the 1.0ml group. Few apoptotic cells were found in the control group. Conclusion Xiaozhengtang can inhibit cell proliferation and promote apoptosis in the graft endometrium.
