摘要
建立了HPLC法测定兔血浆中莲心碱浓度的方法,其浓度在0.125~2.5μg/mI范围内线性关系良好(r=0.9997),平均回收率日内、日间均大于99%,用本法测定静注后兔体内血药浓度的经时变化,结果符合开放性二室模型,其主要药物动力学参数为:t1/2α=8.303min;t1/2β=129.961min;AUC=132.671μg·min·ml;CL=0.0452ml·min^(-1);Vc=2.768L·kg^(-1)。
A method for the determination of plasma concontration of liensinine with HPLC and pharmacokinetic study of liensinine in rabbits were reported. Assay linearity was shown over the range of 0.125~2.5 μg/ml serum with regression coefficient of 0.9-997. The mean recoveries of liensinine in plasma (within-day and between-days) were all more than 99% of the dose and the coefficients of variation were less than 5%. After a single intravenous injection of liensinine 6mg/kg to rabait. The pharmacokinetic characteristics were found to fit a twocompartment open model. Important parameters were: t1/2 α=8.303min tl/2β=129.960minAUC=132.67μg. min/ml CL=0.045 L/min Vc=2.768 L/kg.
