摘要
胸腺素α1是由28个氨基酸残基组成,N端乙酰化的生物活性多肽。采用固相合成方法,以Fmoc氨基酸为原料,经1 氧 3 双二甲胺羧基苯骈三氮唑四氟化硼盐/1 羧基苯并三氮唑(HBTU/HOBt)缩合,三氟乙酸 乙二硫醇 水脱保护,并从Wang树脂上切割下来,粗肽得率为60%,经RP HPLC纯化,获得纯度在98%以上的目标肽。其ESI MS和氨基酸组分分析结果与理论值相符。
Thymosin α_1 was synthesized with solid-phase peptide synthesis on Wang resin, using Fmoc/tBu-HBTU/HOBt activation system. After cleaved with TFA-EDT-H_2O, the crude peptide yield was 60%.After purification with semi preparative RP-HPLC, the purity was found more than 98%. It was identifide on ESI-MS and amino acid analysis.
