血清胆汁酸测定在黄疸鉴别中的应用

摘要 目的 建立高效液相色谱法 (HPLC)测定血清中胆汁酸亚组份的方法及探讨几种黄疸患者的胆汁酸各组份水平的变化。方法 建立HPLC法测定胆汁酸亚组份的方法 ,对所建立的方法进行精密度、准确度、灵敏度及线性范围的评价。用所建立的HPLC法分离、测定几种黄疸患者血清中的 6种胆汁酸并进行比较。结果 HPLC法测定胆汁酸各组份的最低检测限为 3 12 5 μmol L ;在浓度为 3 5~ 10 0 μmol L内 ,其日内变异小于 10 % ,日间变异小于 15 % (除TLCA外 ) ,各胆汁酸的回收率在 93 3%~ 10 6 1%之间 (除TLCA外 )。几种黄疸患者的胆汁酸均高于正常对照组。结论 所建立的HPLC法适用于临床血清胆汁酸的分析。 Objective To build a method to determine bile acids of subsection by HPLC(high performance liquid chromatography) and to investigate bile acids of subsection in serum of patients with jaundice.Methods To build a method to determine bile acids of subsection and to determine the precision, accuracy, sensitivity and linear limitation of the method and to determine six bile acids in serum of patients with jaundice.Results The limitation of determination was 3 125 μmol/L. Coefficients of variation were less than 10% within day and less than 15% day to day for the concentration(3 5~100 μmol/L)of bile acids and recoveries were 93 3%~106 1% except TLCA. The concentrations of bile acids in serum of patients with jaundice were significantly higher than those of the control group( P <0 05).Conclusion The method can meet the need of the determination of six bile acids in serum. The concentrations of bile acids of subsection in sera of patients with jaundice aid to clinical distinguishing diagnosis.
