摘要
目的 观察巯基氧化剂在急性肝损伤时对肿瘤坏死因子α(TNF α)水平的影响。方法 采用半乳糖胺 /脂多糖 (Galn/LPS)所致急性肝损伤模型 ,雄性昆明种小鼠于注射Galn/LPS(每只 4mg/μg)前 1h ,腹腔注射巯基氧化剂二乙基顺丁烯二酸盐 (DEM ) 5mmol/kg ,并分别于Galn/LPS注射 6h后 ,测定血浆ALT活性、TNF α水平和肝脏GSH含量。 结果 DEM预处理可抑制Galn/LPS导致的血浆ALT活性、TNF α水平升高。结论 巯基氧化剂DEM可通过改变肝脏GSH含量的变化 ,影响细胞氧化还原状态 ,抑制LPS导致的TNF α释放 。
Objective To investigate effect of thiol oxidant on tumor necrosis factorα(TNF-α) in acute liver injury. Methods The acute liver injury model ofmale mice was established by LPS plus D-galactosamine(Galn). Male mice were given diethylmaleate (DEM), a thiol oxidant,ip at the dose of 5 mmol/kg 1 h beforeGaln /LPS. Then the levels of ALT,TNF-α in plasma and GSH in liver were detected. Results Pretreatement with DEM could abrogate the high levels of plasma ALT,TNF-α induced by Galn/LPS. Conclusion Redox manipulation throughthiol oxidation DEM may represent a novel approach to prevent Galn/LPS-inducedacute liver injury by inhibition of TNF-α.
