摘要
目的 建立Wistar、SD大鼠及BALB/c小鼠脑胶质瘤动物模型 ,比较其颅内肿瘤生长特点。方法 借助动物立体定向仪 ,将体外培养大鼠C6胶质瘤细胞 (细胞数为 3× 10 5个 ,悬浮于无血清DMEM培养液 6 0 μl中 )分别接种于Wistar、SD大鼠左侧尾状核区 ,BALB/c小鼠左顶枕区 (接种细胞数为 1× 10 3 )。接种后观察不同种实验鼠的生存状态及肿瘤的生长特性 ,分别于接种后 5、15d进行MRl检查。在实验 3d、6d、9d、12d、15d时解剖标本 ,做组织病理学和胶质纤维酸性蛋白(GFAP)免疫组化检查。结果 两种大鼠脑胶质瘤动物模型在组织病理学上接近人脑胶质瘤 .而且具有颅内生长稳定 ,成瘤率高 ,未见颅外转移病灶 ,实验周期短 ,重复性好。而BALB/c小鼠成瘤率76 .6 7%。结论 接种的Wistar、SD两种大鼠脑胶质瘤动物模型 ,其肿瘤生长特性及病理特征与人脑胶质瘤相似。均可作为临床胶质瘤基础研究的理想模型。而BALB/c小鼠则需进一步探索。
Objective To explore the growth of brain glioma, Wistar, SD, BALB/c C6 brain glioma models were established with stereotactic technique.Methods The C6 cells cultured in vitro were stereotaxically implanted into the left caudate nucleus of Wistar, SD rat brain and BALB/c in the left parieto occipital lobe. The concentration of C6 cells was 5×10 9 L -1 free serum DMEM ( the volume of injection 60 μl for rats, 6 μl for mouse). After C6 inoculation, the following step was to use an MRI protocol to evaluate the growth and vascularity of implanted C6 rat gliomas on 5 and 15 day. The living and survival states of animals with glioma were observed too. In order to study the size of tumor, survival time and histopathology of the tumor, the animals with tumors were killed on 3, 6, 9, 12, 15 day after implantation. Tumor was confirmed with HE and staining of glial fibrillary acidic protein(GFAP)immunohistochemistry.Results Rat C6 gliomas resembled histopathological features of human glioma. This kind of model was a more reliant and reproducible one, with 100% yield of intracranial tumor as well as no extracranial growth extension. The tumor was observed in 23 mice (76.67%).Conclusion A rat C6 brain glioma model resembles histopathological features of human glioma , which can be used as a perfect model to study the etiology of glioma. Whereas the model of BALB/c C6 glioma should be explored in the future.
