酸性成纤维细胞生长因子的纯化和生物学特性检测

Purification of Acidic Fibroblast Growth Factor from Bovine Pituitary And Its Biological Characteristics

摘要 采用硫酸胺沉淀,CM—Sephadexc_(50)及heparin—Sepharose层析三步法,从牛脑下垂体中纯化酸性成纤维细胞生长因子(a—FGF),获得量190.22μg/kg组织,其分子量15400,等电点5.1。它可刺激小鼠L_(929)及人HFC成纤维细胞增殖及DNA合成。最大增殖效应所需浓度为9ng/ml,50%最大增殖效应所需浓度为2ng/ml。 Pituitary acidic fibroblast growth (aFGF) was purified by a three-step procedure, including ammonium sulfate precipitation, CM-sephadex c50 ion-exchange chromatography and heparin-sepharose affinity chromatography. The fraction H2 of aFGF was obtained in a yield of 1.87ug/kg of tissue. Its molecular weight is 15400. It is active in stimulating proliferation and promoting DNA synthesis of L929 and HFC. Dose-cells response studies indicate that fraction H2 of aFGF stimulated L929 cells proliferation maximally at a concentration of 9ng/ml, while half-maximal response ocurred with 2ng/ml.
