弓形虫P30乳酸球菌口服疫苗诱导小鼠的细胞免疫反应及抗体生成的动态过程 被引量:8

摘要 目的 观察弓形虫P30乳酸球菌口服疫苗诱导小鼠产生的细胞免疫效果和抗体IgG的动态发生过程。方法 口服免疫BALB/c小鼠 ,一个月后 ,取鼠脾细胞作ConA刺激淋转试验 (MTT法 )及T细胞亚群的测定 ;不同免疫时间段内 ,收集实验鼠血清 ,ELISA法检测抗体水平的变化。结果 P30乳酸球菌口服免疫组脾淋巴细胞ConA刺激后增殖能力比两对照组显著提高 ,CD+ 4 和CD+ 8的百分数均明显升高 ;第 12周 (即最后一次免疫结束一个月 ) ,P30乳酸球菌口服免疫组检测到特异性抗体IgG。结论 P30乳酸球菌口服疫苗有效地激发了小鼠细胞免疫反应 ,特异性抗体IgG在免疫结束一个月后生成明显。 Objective To study the cellular response and the kinetics of development of antibody IgG in mice by oral vaccination of recombinant Lactococcus lactis expressing P30 of Toxoplasma gondii.Methods After one month of oral vaccination,the proliferation of mice spleen lymphocytes were detected by MTT assay and the number of CD+ 4 and CD+ 8T cell were achieved by flow cytometer assay;sera were obtained at different immune time,and were tested for the kinetics of development of antibody IgG.Results The proliferation of spleen lymphocyte from mice immunized with L.lactis/P30 was enhanced significantly after stimulation with ConA,the number of both CD+ 4and CD+ 8T cell increased compared with two control groups;specific antibody IgG was obviously detected in mice orally immunized with L.lactis/P30 in twelve weeks(i.e.one month after the last immunization).Conclusion Vigorous cellular immune response was observed by oral vaccination of recombinant L.lactis /P30 in mice;specific antibody IgG against the recombinant P30 protein begin to generate significantly in one month after immunization.
