喷昔洛韦对单纯疱疹病毒Ⅰ型和Ⅱ型的抑制作用 被引量:4

摘要 目的 评估喷昔洛韦钠盐在体内外对HSV Ⅰ和HSV Ⅱ感染的抑制作用。方法 在组织培养中用CPE抑制实验测定喷昔洛韦钠盐对HSV Ⅰ和HSV Ⅱ所致vero细胞病变的抑制作用 ;在BALB/c小鼠中测定其对HSV Ⅰ感染BALB/c小鼠所致耳部皮损的治疗作用。结果 喷昔洛韦钠盐能有效抑制 10 0TCID50 HSV Ⅰ (sm44株 )和HSV Ⅱ (3 3 3株 )感染vero细胞所产生的细胞病变作用 ,半数有效剂量 (IC50 )分别为 1.95 μg/ml和 3 .90 μg/ml ;使HSV Ⅰ病毒感染性滴度下降 99%的药物浓度 (IC99)为 2 .40 μg/ml。在HSV Ⅰ (sm44)感染BALB/c小鼠中 ,喷昔洛韦钠盐大剂量组 (2 5 0mg/kg·d)、中剂量组 (10 0mg/kg·d)、小剂量组 (5 0mg/kg·d)均能明显降低HSV Ⅰ感染BALB/c小鼠所致耳部皮损作用。 结论 喷昔洛韦钠盐在体内外具有抗HSV Ⅰ和HSV Ⅱ感染的作用。 Objective To evaluate antiviral activity of penciclovir against HSV-Ⅰ and HSV-Ⅱ infection in vitro in vivo.Methods The CPE inhibitions of vero cells infected by HSV-Ⅰ and HSV-Ⅱ were assayed by CPE inhibitory experiments;The treatment effect on the lesion of ear pinna of BALB/c mice infected by HSV-Ⅰ were determined for evaluating antiviral activity of penciclovir.Results Penciclovir could effectively inhibit CPE of vero cell infected by HSV-Ⅰ,and the IC 50 for penciclovir against HSV-Ⅰ and HSV-Ⅱin vero cell were 1.95 ug/ml and 3.90 ug/ml,respectively.The concentration of required to inhibit the HSV-Ⅰ yield by 99%(IC 99 ) was 2.40 ug/ml.Treatment with the compound(250 mg/kg·d,100 mg/kg·d and 50 mg/kg·d groups) was highly effective in reducing the lesion score of BALB/c mice.Conclusion Penciclovir has antiviral activity against HSV-Ⅰ and HSV-Ⅱ in vitro and in vovo.
