摘要
目的:探讨姜黄、郁金和莪术等3种姜黄属中药抗氧化、降血脂、抗肿瘤等作用的主要活性成分姜黄色素的提取、分离方法及其紫外吸收特点。方法:采用有机溶媒法提取、分离其中的姜黄色素成分,然后用柱色谱法获取3种色素单体;用薄层层析法对各成分进行纯度鉴定,同时应用紫外光谱扫描分析色素成分的紫外吸收特点。结果:①姜黄色素是3种姜黄属中药的共有成分,以姜黄含量为最高。②乙醚脱脂效果明显优于石油醚。③柱色谱法可获取纯度很高的多种色素单体。④3种姜黄色素单体的紫外吸收图谱基本相同,其特征性吸收峰均位于420~425nm处。结论:有机溶媒法与柱色谱法相结合是提取姜黄抗氧化成分-色素及其单体成分的有效方法;3种主要色素成分相似的紫外吸收特点。
AIM:To analyze extraction and isolation methods and character of ultraviolet visible absorption of the main active component, curcumin which has the functions of anti oxidation,blood lipid reducing and anti tumor etc. in three species of curcuma herbs,turmeric,aromatic turmeric root tuber and zedoary. METHODS: Curcumin was extracted and isolated by using organic solvent,then column chromatography was used to get curcumin 1 (curcumin), curcumin 2(demethoxycurcumin) and curcumin 3 (bisdemethoxycurcumin) from curcuminoids.Thin layer chromatography(TLC) was performed to evaluate purity of all the components, meanwhile ultraviolet visible spectrophotometry was used to analyze character of ultraviolet visible absorption of segment components we had got. RESULTS:①The mutual component of the three species of curcuma herbs was curcumin,the highest content of which was the tumeric. ②Degreasing effect of ether was obviously better than that of petroleum ether. ③A great deal of pigment monomer in high purity could be got by using column chromatography.④Ultraviolet visible absorption spectrum of curcumin 1, curcumin 2 and curcumin 3 were almost the same and their characteristic absorption peak had a height of 420-425 nm.
