东方田鼠血清及其不同部分对日本血吸虫童虫的体外杀伤作用 被引量:17

摘要 目的 对东方田鼠血清及其不同部分对日本血吸虫童虫的体外杀伤的可能机制进行初步探讨。方法 将血清初步分离成含蛋白部分及分子量小于 2 0 k Da的小分子部分 ,并分别观察了东方田鼠全血清、去补体血清、蛋白及小分子血清成分对体外培养童虫的杀伤 ,此外 ,还观察了蜕皮素对东方田鼠血清杀伤作用的影响。结果 东方田鼠热灭活去补体血清对童虫的杀伤显著低于正常血清 ;感染及正常东方田鼠血清小分子在第 4 8h及 72 h表现出一定杀伤作用 (30 % ) ,而血清蛋白在2 4 h就有显著杀伤 ,但杀伤作用略低于全血清 ;蜕皮素对东方田鼠血清的杀伤无明显影响。结论 补体在东方田鼠血清杀伤机制中有重要作用。血清蛋白为体外杀伤的主要成分 。 ObjectiveTo explore the poss ib le mechanisms of killing effects of serum from Microtus fortis to Schistosoma japonicum schisto somula in vitro. MethodsSerum was separated into protein fraction and <20 k Da small molecule fraction. The killing effects to schistosomula with fresh and h eat inactivated seru m from Microtus fortis(MFs), and the serum fract ions of protein and small molecu lar weight(MW) part were observed respectively. In addition, the effect of e cdysone on the k illing activity was also observed. ResultsMFs lost its killing effect when it was heat inactivated. After incubatio n with serum component of small MW for 48 h or 72 h, 30% of organisms were found t o be dead. The serum protein had an obvious killing effect at the 24 h, however, a little lower than that of complete serum. Ecdysone showed no effect on the kil ling activity. ConclusionComplement has significant effect on killing schistosomula in vitro[WT6BZ ]. Serum protein is the major component in the killing effect, and the small MW part is also involved in the killing.
