脐血T淋巴细胞亚群分布特点及其临床意义探讨 被引量:3

摘要 目的 比较脐血、骨髓及外周血中T淋巴细胞亚群的分布特点 ,探讨脐血移植后免疫重建延迟的发生机制。方法 应用流式细胞仪分别检测脐血、骨髓及外周血中CD3+ 、CD3+ CD5 + 、CD4 + 、CD4 + CD2 8+ 、CD8+ 和CD8+ CD2 8+ 细胞的含量。结果 CD3+ 、CD4 + 及CD8+ 细胞数在脐血、骨髓及外周血中无显著差异 ,P >0 .0 5。脐血中CD3+ CD5 + 、CD4 + CD2 8+ 及CD8+ CD2 8+ 细胞数显著低于骨髓及外周血 ,P <0 .0 5。骨髓与外周血中各T淋巴细胞亚群测定无显著差异 ,P >0 .0 5。结论 脐血中成熟的T淋巴细胞 (CD3+ CD5 + )及细胞毒T淋巴细胞 (CD4 + CD2 8+ 、CD8+ CD2 8+ )的数量显著低于骨髓及外周血 。 Objective To study the mechanism of delayed immune reconstitution after cord blood transplantation by compared with the characteristics of distribution of T lymphocyte subsets in cord blood,bone marrow and peripheral blood.Methods The amounts of CD3 +,CD3 +CD5 +,CD4 +,CD4 +CD28 +,CD8 + and CD8 +CD28 + cells in cord blood,bone marrow and peripheral blood were respectively measured with flow cytometry.Results There was no marked change on the amounts of CD3 +,CD4 + and CD8 +cells in cord blood,bone marrow and peripheral blood( P >0.05 ).The amounts of CD3 +CD5 +,CD4 +CD28 + and CD8 +CD28 + cells in cord blood were significantly decreased compared with bone marrow and peripheral blood( P <0.05 ).The amounts of each T lymphocyte subsets in bone marrow were not significantly changed compared with peripheral blood( P >0.05 ).Conclusion The amounts of mature T lymphocyte(CD3 +CD5 +) and cytotoxic T lymphocyte(CD4 +CD28 +,CD8 +CD28 +) in cord blood were significantly decreased compared with bone marrow and peripheral blood.Maybe this is the main cause of delayed immune reconstitution after cord blood transplantation.
