摘要
目的 研究不同材质导尿管对铜绿假单胞菌生物膜形成的影响及不同物理方法对不同生长时期生物膜的清除效应。方法 体外用平板法 ,经过 7d孵育在 4种导尿管材质膜片上均培养出生物膜模型。采用乳胶镀膜、硅胶镀膜、纯硅胶和红橡胶等不同材质导尿管的膜片制备铜绿假单胞菌生物膜 ,通过菌落计数法了解粘附于导尿管材生物膜内细菌菌落数量与材质的关系 ;用冲洗、冲洗 +涡旋振荡、冲洗 +超声振荡 3种方法处理附有不同生长时期生物膜的导尿管 ,通过菌落计数法了解 3种方法对不同生长时期生物膜的清除效应。结果 以乳胶镀膜和硅胶镀膜所分离出的生物膜内细菌最多 ,分别为 1 .91× 1 0 7CFU/cm3 和 1 .2 0× 1 0 7CFU/cm3 ;以纯硅胶上所分离的生物膜内细菌最少 ,菌落数为 2 .7× 1 0 5CFU/m3 ,红橡胶上所分离的菌落数稍高于纯硅胶 ,为6 .5× 1 0 6CFU/cm3 ,四者两两比较 ,均有统计学差异 (P <0 .0 0 1 )。不同处理方法结果表明 ,在生物膜形成的初期 (2d) ,3种处理方法具有相同清除效应 ;但当生物膜趋于成熟后 ,单用冲洗方法难于将粘附细菌洗脱下来 ,3种方法的效应强弱顺序为 :冲洗 +超声 >冲洗 +涡旋振荡 >冲洗。结论 不同材质导尿管与铜绿假单胞菌形成生物膜的数量有一定关系 。
Objective To investigate the effect of different catheter materials against formation of P .aeruginosa biofilm,and to learn the eradicative efficacy of various treatment to different growth period of biofilm.Methods P.aeruginosa biofilms were prepared with four kinds of catheter, which were made of latex, siliconized latex, pure silicone and rubber ,incubated with P.aeruginosa for 7 days, and the viable bacteria counts of biofilm were performed by serially diluted method. In addition, three ways including washing, washing and vortex agitation, washing and ultrasound were employed to treat P. aeruginosa biofilm in different growth period.Results The viable bacterial counts of biofilm adhered to latex and siliconized latex, were 1.91×10 7CFU/cm 3 and1.20×10 7CFU/cm 3,respectively ,compared with 2.7×10 5CFU/cm 3 of biofilm adhered to pure silicone, The counts of biofilm adhered to rubber were slightly higher than pure silicone ,which is 6.5×10 6CFU/cm 3.Statistical difference consisted in the counts of biofilm adhered to four kinds of materials.The eradicative efficacy of various treatment showed, that three ways had similar efficacy of eradicate the young P. Aeruginosa biofilm, but washing was difficult to eradicated the mature biofilm from catheter,compared with the strongest eradication of wash plus ultrasound .Conclusion There was a certain relationship between biomaterial and formation of P. aeruginosa, catheters made of latex and siliconized latex were easily adhered to bacteria and formation of P.aeruginosa, compared with pure silicone and rubber; washing was effective methods to eradicate young biofilm adhered to catheters.
