乳腺组织中钠/碘转运体基因及蛋白的表达与乳腺疾病发生的关系 被引量:4

Relationship of sodium-iodide symporter gene and protein expression in breast tissue with the occurrence of breast disease

摘要 目的:研究正常和良/恶性乳腺肿瘤组织中钠-碘转运体(sodium-iodidesymporter,NIS)基因和蛋白的表达情况,探讨NIS与乳腺疾病之间的关系。方法:乳腺组织取自4例乳腺纤维瘤患者,3例乳腺小叶增生患者,8例乳腺癌患者,5例癌旁正常乳腺组织,运用半定量反转录聚合酶链反应(RT-PCR)和免疫组织化学技术检测NIS的基因与蛋白表达情况。结果:癌旁正常乳腺组织和良性乳腺肿瘤组织未检出NIS基因和蛋白的表达,而乳腺癌组织NIS基因均见NIS基因表达,且NIS蛋白主要表达在胞浆中。结论:正常乳腺组织中未检测到NIS基因,而乳腺癌组织NIS基因表达明显增加,而NIS蛋白翻译和翻译后水平的缺陷可能是乳腺癌组织摄碘率降低的原因之一。 AIM:To study the expression of sodium-iodide symporter(NIS) gene mRNA and pro tein in normal and benign or malignant breast tissue so as to discuss the relati onship between NIS and breast disease.METHODS:Breast tissues were dissected from 4 patients with breast fibroid,3 wi th mammary lobular hyperplasia, 8 with breast cancer and 5 with normal breast ti ssue.The expression of NIS gene mRNA and protein were detected by using RT-PCR and immunohistochemistry.RESULTS:The expressions of NIS gene mRNA or protein were not found in benign b reast nodule and normal breast tissue,while NIS gene mRNA was found in the breas t cancer tissues,and mainly expressed in cytoplasm.CONCLUSION:Expressions of gene mRNA or protein of NIS are not detected in beni gn breast nodule and normal breast tissue,but increased markedly in breast cance r tissue.The defect of NIS mRNA transcription or post-transcription may be one of the reasons for the decrease of uptake rate of iodide in breast cancer tissue .
