摘要
目的:探索经验方丹芪注射液经关节腔注射后对创伤性关节炎(post-traumaticarthritis,TA)家兔模型血清、关节滑液中透明质酸(HA)含量的影响。方法:将40只家兔中的30只的右膝关节行半月板切除术以建立TA动物模型,随机数字表法分成丹芪组、透明质酸钠组及模型组,每组10只,然后分别经关节腔注射的途径予以丹芪注射液,透明质酸钠及不给药。分别于治疗前后观察动物右膝关节肿胀变化,同时抽取动物血液标本及关节冲洗液检测HA的含量(放免法)。结果:在造模手术后13周时造模成功。经6周的治疗(每3d1次)后,动物关节的肿胀程度、血清中HA的含量均有不同程度的下降,其中丹芪组优于透明质酸钠组(P<0.05),而关节滑液中HA含量变化两组间比较差异无显著性意义(P>0.05)。结论:半月板切除可以建立TA动物模型,丹芪注射液能够改变TA家兔模型血清及关节液中HA的含量,从而证明丹芪注射液经关节腔注射后能够在一定程度上防止或改变创伤后关节的退行性病变。
AIM:To explore the effects of danqi injection by intraarticular administration on the content of hyaluronic acid(HA) in serum and synovia fluid of rabbit knee with post-traumatic arthritis(TA). METHODS:Forty rabbits were randomly divided into model group(n=30) and normal control group(n=10).After TA animal models were set up the by performing menisce ctomy to the right knee joints of rabbits in the model group,the model group wer e subdivided randomly into danqi group,sodium hyaluronate(SH) group and model co ntrol group(10 rabbits in each group). Then the subdivided groups were injected intraarticularly with danqi injection,SH or without injection.The changes of rig ht knee joints were observed pre- and post-injection.The circulating blood an d synovia lavage fluid samples were taken in order to measure the content of HA( radioimmunoassay). RESULTS:The modeling succeeded 13 weeks after operation of modeling.13 weeks l ater surgery, The content of HA in synovia fluid from the model was lower than t he normal animals,whereas that of HA in serum from the model was higher.After 6 -week treatment(once every 3 days),the swelling of joints and the content of HA in the blood serum decreased at different degrees,and changes of them in the da nqi group were superior to those in the SH group(P< 0.05),while the content of H A in synovia fluid had insignificant difference between the two groups(P >0.05). CONCLUSION:The TA animal model can be set up by meniscectmy.Danqi injection ca n change the contents of HA in the serum and the synovia fluid of rabbits,which indicates that intraarticular administration of danqi injection can prevent or c hange the degeneration of TA in a certain extent.
