细胞黏附分子在骨肉瘤中的表达及其与肿瘤侵袭转移、预后的关系(英文)

Expressions of cellular adhesive molecule in osteosarcoma and its relationships with tumorous attack, metastasis and prognosis

背景:骨肉瘤的术后转移是造成治疗失败的主要原因,而发生机制目前尚不明确。有研究证明黏附机制的失调在肿瘤的转移中起着重要的作用。目的:探讨CD44v6和P选择素在骨肉瘤组织中的表达特征及其与骨肉瘤术后侵袭转移的相关性。设计:采用非随机对照设计。地点和对象:以本院1994/2002间手术切除34例骨肉瘤标本和同期收治的10例骨软骨瘤作为研究对象。干预:采用流式细胞术对34例骨肉瘤组织和10例骨软骨瘤组织中的P选择素和CD44v6的表达进行荧光免疫检测。主要观察指标:肿瘤组织中CD44v6及P选择素阳性表达率。结果:在骨肉瘤组织中,CD44v6及P选择素阳性表达率明显高于骨软骨瘤;不同外科分期间的CD44v6和P选择素表达,差异有显著性意义;CD44v6和P选择素表达与骨肉瘤肺转移有明显相关性。结论:P选择素和CD44v6与骨肉瘤的侵袭、转移相关。可作为骨肿瘤恶性表型和预测骨肉瘤转移潜能的指标。

BACKGROUND:The main reason for the therapeutic failure in osteosarcoma is its post-surgery metastasis.However, the pathogenesis is still not well established .Some researches demonstrated that the imbalance of adhesive mechanism played an important role in tumorous metastasis.OBJECTIVE:To discuss the expressive characteristics of CD44v6 and P lectin and their correlations to post-surgery metastasis in osteosarcoma.DESIGN:A non-random control study was performed.SETTING and PARTICIPANTS:A total of 34 osteosarcoma specimens removed by surge ries between 1994 to 2002 as well as 10 osteochondroma patients admitted at same period in General Hospital of Jinan Military Region were selected.INTERVENTIONS:The expression of P lectin and CD44v6 in 34 osteosarcoma tissues and 10 osteochondroma tissues were assayed using immunofluorescence technique b y flow cytometry.MAIN OUTCOME MEASURES:Positive expressive rates of CD44v6 and P lectin in tumo r tissues.RESULTS:The positive expressive rates of CD44v6 and P lectin were distinctly h igher in osteosarcoma tissues than that of osteochondroma tissues.There were sig nificances in the expressions of CD44v6 and P lectin among different surgical st ages.There were significant correlations between the expressions of CD44v6 and P lectin, and the lung metastasis of osteosarcoma.CONCLUSION:P lectin and CD44v6 correlate with the attack and metastasis of ost eosarcoma, which can be used as a malicious phenotype of bone tumors and an inde x in predicting the potential metastasis ability of osteosarcoma.
