摘要 目的:研究人垂体腺瘤中Myc/Max/Mxi1网络的异常。方法:58例垂体腺瘤分组:非侵袭性与侵袭性垂体腺瘤;小腺瘤与大腺瘤;复发性与非复发性垂体腺瘤。采用免疫组化ABC法检测不同分组中的Myc,Mxil和Max的表达。结果:垂体腺瘤中Myc的过表达率45%,Mxil和Max的缺失率为90%和86%。Myc阳性率及表达强度在侵袭组与非侵袭组,复发组与非复发组中均差异有显著性意义。58例肿瘤组织中31例有1种以上的因子异常,而正常脑组织中仅1例异常。结论:Myc/Max/Mxi1网络异常与垂体腺瘤的发生和发展关系密切,Myc/Max/Mxi1网络系统具有调节垂体肿瘤细胞的增殖与分化功能。Myc的表达是预测垂体腺瘤生物学行为及预后的良好指标。 AIM:To investigate the relationship between the expression of Myc,Max and Mxil proteins and human pituitary adenomas(hPA).METHODS:Totally 58 PA were classified as invasion and non-invasion PA,microad enomas and macroadenomas, recurrence and non-recurrence PA.Immunohistochemical assay was used to determine protein expression of c-Myc,Mxil and Max. RESULTS:Over expressed Myc protein was found in 45% of the 58 PA, and Mxil a nd Max failed to express in 90% and 86% of the PA respectively. The positive expressive ratio and level were statistically different between invasions and n on-invasion group and between recurrence and non-recurrence group(P< 0.05).Of the 58 eases of PA, it was found that 31 cases had at least one protein abnormal ly expressed,comparing with only one case in 10 normal brain tissues.CONCLUSION:The abnormality of Myc/Max/Mxil network may be closely correlated w ith the occurrence and progress of PA,and play an important role in regulating c ell proliferation and differentiation in PA.The expression of Myc protein can be used as a promising index in PA prognosis.
