脑出血后c-jun基因表达的研究 被引量:2

摘要 目的 探讨c jun基因在血肿周围缺血带的表达与神经元生存和恢复及预后的相关性。方法 根据入院时的格拉斯哥 (glasgowcomascale,GCS)评分情况 ,对 30例脑出血患者分为GCS≥ 8分组和GCS <8分组 ;按治疗后两周时病情转归将患者分为死亡组和存活组 ;另取非脑系科疾病标本 10例作为对照组。用血肿清除术或血肿碎吸术中所带出的血肿周围组织标本行地高辛标记探针原位杂交 ,检测c junmRNA的表达水平 ,以多功能彩色病理图像分析系统进行分析。结果 脑出血组c junmRNA阳性神经细胞百分率和平均吸光度值均显著高于对照组 (分别为5 .4 3± 1.2 0及 3.0 0± 0 .35 ,P <0 .0 1) ;c junmRNA的阳性神经细胞百分率和平均吸光度值在死亡组的表达高于存活组。结论 脑出血的缺血性损伤诱导c jun基因表达 ,出血量越大 ,病情越重 ,表达越明显。 Objective Through analysis of the relationship between the expression of c-jun in the surrounding of hematoma and the survival of neurons and the prognosis,to provide a basis for precise evaluation of the extent of brain injury and prognosis of cerebral hemorrhage.Methods According to the Glasgow-Pittsburgh coma scale(GCS) value of the patients, 30 cases of cerebral hemorrhage were divided into two groups:GCS≥8 group and GCS<8 group.Based on the status of two-week treatment,the cases were divided into two groups:the death group and the survival group. The expression of c-jun gene in the local neural tissue was in vestigated by in situ hybridization and the results were analyzed on colour multi-functional image analysis system.Results The positive rate and ND of c-jun mRNA in the cerebral hemorrhage group were higher greatly than those in the control group (P<0.01). The expression of c-jun mRNA in the death group and GCS<8 group was higher than that in the survival group and GCS≥8 group (P<0.05).Conclusion It suggests that the expression of immediate early c-jun gene correlates with tissue injury in cerebral hemorrhage.
