摘要 目的 :观察酸性成纤维细胞生长因子 (aFGF)和表皮生长因子 (EGF)对内侧副韧带 (MCL)和前十字韧带 (ACL)细胞增殖行为的影响。方法 :培养 1 0周龄新西兰白兔内侧副韧带和前十字韧带细胞 ,在培养液中分别加入aFGF和EGF ,以XTT方法测定细胞的增殖行为。结果 :aFGF在 1ng/ml时即对两种细胞具有显著的促进增殖作用 ,其浓度达 50ng/ml时 ,对MCL细胞的促进作用最大 ,达1 0 0ng/ml时对ACL细胞的促进作用最大。EGF在 0 .78ng/ml时即对MCL细胞有显著的促增殖作用 ,在 1 .56ng/ml时始对ACL细胞有显著的促增殖作用 ,其浓度达 3 .1 2 5ng/ml时对 2种细胞的促进作用最大。aFGF和EGF在超过其最佳浓度后 ,随浓度升高促进作用均下降。结论 :aFGF和EGF可以促进韧带成纤维细胞增殖。 Objective:To observe the effects of acid fibroblast growth factor(aFGF) and epidermal growth factor(EGF) on the proliferation of the cells isolated from medial collateral ligament(MCL) and anterior cruciate ligament(ACL) and ACL in vitro.Methods: The MCL and ACL cells of ten-week-old skeletally immature New Zealand white rabbit were cultured,while aFGF and EGF were added to the cell culture media,the cellular proliferation was assayed using XTT method.Results: The addition of 1ng/ml aFGF enhanced the proliferation of both MCL and ACL cells significantly,this enhancement of proliferation of MCL cells was maximal in the concentration of 50ng/ml and it achieved maximal enhancement when ACL cells were cultured in aFGF at 100ng/ml.The addition of 0.78ng/ml EGF enhanced the proliferation of MCL cells significantly and it enhanced the proliferation of ACL cells at 50ng/ml,the enhancement of proliferation of MCL and ACL cells was maximal in the concentration of 3.125ng/ml.The efficacy of aFGF and EGF declined gradually when they present at concentrations greater than the dose with the greatest efficacy.Conlusion: It is evident that aFGF and EGF can stimulate the proliferation of the ligamentous cells.
