H_(22)微波瘤苗免疫小鼠抑瘤及免疫效果的研究

摘要 目的 :研究H2 2 微波瘤苗免疫小鼠的抑瘤及免疫效果。方法 :用微波处理H2 2 瘤株制备瘤苗 ,接种昆明小鼠右后肢外侧皮下 0 2mL ,共接种 3次 ,每次间隔 7d ;对照组接种生理盐水。于末次接种后第 7天每只小鼠左腋下皮下接种H2 2 瘤株细胞 2× 10 6 。接种瘤株后第 2 1天处死小鼠 ,测量瘤块的体积和重量 ,计算体积抑瘤率和重量抑瘤率 ,并进行血液淋巴细胞亚群分析和体内淋巴细胞转化实验。结果 :瘤苗组瘤块体积平均为 2 86cm3,明显小于对照组的 8 0 2cm3,差异有极显著意义 ,t=5 711,P =0 0 0 0 ,体积抑瘤率为 6 4 34%。瘤苗组瘤块重量平均为 2 5 5g ,明显小于对照组的 6 35g ,差异有极显著意义 ,t′ =5 0 5 5 ,P =0 0 0 0 ,体积抑瘤率为 5 9 84 %。瘤苗组CD3+ ,t =3 2 78,P =0 0 0 3、CD4 + ,t =2 5 48,P =0 0 17、CD8+ ,t =2 0 6 8,P =0 0 4 8和CD4 + /CD8+ 比值 ,t =2 116 ,P =0 0 4 3,均以瘤苗组高于对照组 ,差异有显著意义或极显著意义。淋巴细胞转化率瘤苗组亦高于对照组 ,差异有极显著意义 ,t=4 976 ,P =0 0 0 0。结论 :H2 2 微波瘤苗免疫小鼠对H2 2 瘤株具有明显抑瘤作用。 Objective To study the antitumor and immunological effects of the immunization of microwave treated H 22 tumor vaccine (MTTV H 22 ) on mice.Methods The MTTV H 22 was made up of H 22 tumor cells treated with microwave.The mice were immunized on their right back legs (0 2 mL) with MTTV H 22 3 times with the intervals of 7 days.The control group were treated with the N S instead of MTTV H 22 .Seven days after the last immunization,the H 22 tumor cells were inoculated to the left axillary of the mice (2×10 6 cells/mouse).Twenty one days after the inoculation,the mice were sacrificed and the sizes and the weights of the tumor were measured.The lymphocyte subgroup analysis and examination of the lymphocyte tansformation test were also performed.Results The average tumor size in the MTTV H 22 group (2 86 cm 3) was obviously smaller than that in control group (8 02 cm 3) with the high significant difference, t =5 711, P =0 000.The average tumor weight in the MTTV H 22 group (2 55 g) was obviously lighter than that in the control group (6 35 g) with the high significant difference, t′ =5 055, P =0 000.The antitumor rate was 64 34% in size and 59 84% in weight.The subgroup of CD 3 +, t =3 278, P =0 003,CD 4 +, t =2 548, P =0 017,CD 8 +, t =2 068, P =0 048,lymphocyte and the rate of CD 4 +/CD 8 +, t =2 116, P =0 043,were all higher in the MTTV H 22 group than in the control group with the statistical differences.The rate of lymphocyte transformation were also higher in the MTTV H 22 group than in the control group with the high significant difference, t =4 976, P =0 000.Conclusions The immunization of microwave treated H 22 tumor vaccine (MTTV H 22 ) on mice shows obvious antitumor effect.
