摘要
目的 :探讨丁酸钠 (NaB)对人子宫内膜癌细胞系HHUA细胞体外增殖的影响及机制。方法 :不同浓度的NaB作用于体外培养的人子宫内膜癌细胞系HHUA ,通过绘制细胞生长曲线观察NaB对细胞生长的抑制作用 ;用流式细胞仪定量分析细胞周期分布 ;用WesternBlot分析p2 1、p5 3、pRb和E2F1蛋白表达。结果 :NaB能诱导HHUA细胞周期G1期阻滞 ,上调p2 1蛋白表达 ,促进pRb去磷酸化 ,下调E2F1和p5 3蛋白表达。结论 :NaB能抑制HHUA细胞体外生长 ,其诱导细胞周期G1期阻滞可能与p2 1等细胞周期调节蛋白的表达改变有关。
Objective To study the effect of sodium butyrate(NaB) on proliferation of human endometrial carcinoma cell line HHUA cells and its mechanism.Methods Human endometrial carcinoma cell line HHUA cells were cultured in vitro and treated with different concentrations of NaB.The growth suppressing effect of NaB on HHUA cells was observed by drawing growth curves.Cell cycle distribution was quantified by using flow cyto metric analysis.Western blot analysis was carried out to determine the expression of p21,p53,pRb and E2F1 proteins.Results NaB induced G1 phase arrest in HHUA cells.NaB upregulated p21 protein expression,promoted pRb dephosphorylation and downregulated E2F1 and p53 proteins expression.Conclusions NaB can inhibit growth of HHUA cells in vitro.Changes in p21 and other cell cycle regulation associated proteins may be the mechanism of G1 phase arrest.
