摘要
目的 寻找简便快速的从全血中分离高质量DNA的方法。方法 分别用异硫氰酸胍 硅胶法和经典的酚 氯仿法从全血中提取DNA并进行比较。结果 异硫氰酸胍 硅胶法提取DNA平均含量 (3.70 μg/ 10 0 μl全血 )高于酚 氯仿法 (2 .99μg/ 10 0 μl全血 ) ,P <0 .0 5 ,纯度无显著差异。 2法抽提的DNA琼脂糖凝胶电泳结果及PCR和限制性酶切反应的效果无差异 ,但异硫氰酸胍 硅胶法更为简便 ,所需时间不到 1h。结论 异硫氰酸胍 硅胶法提取全血中DNA简单。
Objective To search a simple,rapid method for extracting DNA from whole blood.Methods GuSCN-silica gels procedure was used to isolate DNA from whole blood and compared with conventional phenol-chloroform extraction method.Results The characteristics of quality (e.g.,purity,bands in electrophoresis,effect of restrict endonuclease and results of PCR)of DNA obtained by the GuSCN-silica gels method was comparable to those obtained by phenol-chloroform extraction,but the quantity of DNA obtained by GuSCN-silica gels method was more than that of phenol-chloroform extraction.The DNA extracted by the two methods has been successfully used for PCR and enzyme digestion,in which the results were completely coincident.Conclusion The GuSCN-silica gels procedure is a simple,rapid and efficient method for extra ction of genomic DNA from whole blood.
