摘要 目的 检测戊型肝炎病毒 (HEV)Ⅳ型开放阅读框架 2 (ORF2 )编码蛋白N 端表位的抗原性 ,寻找良好的抗原肽 ,以便用于HEV抗体测。方法 根据HEVORF2编码蛋白N 端中含有的抗原表位 ,从HEV中国株蛋白的氨基酸序列中选取 2 5~ 38位序列氨基酸 ,用固相化学合成法合成抗原肽 ,并用ELISA检测抗原性。结果 HEVⅣORF2编码蛋白N 端氨基酸顺序为2 5~ 38的抗原肽与急性期戊型肝炎病人血清有较高的反应性 ,与Genelab诊断试剂盒的符合率达 93.8%。结论 为提高HEV抗体的检出率 ,在包被抗原中应含有HEVⅣ型ORF2编码蛋白N Objective To search a better antigen peptide for the preparation of HEV EIA diagnostic kit.Methods Based on the antigen epitopes included in the N-terminal portion of peptide encoded by HEV ORF2,the N-terminal amino acid sequence 25 to 38 of Chinese strain HEV protein was selected to synthesize the antigenic peptide.The peptide was synthesized in terms of the method of solid phase peptides synthesis (SPPS).Its antigenicity was identified by ELISA.Results The synthetic peptide has higher sensitivity in the detection of IgG-type antibodies in serum from acute hepatitis E patients.Of the results 93.8% was coincidental to Genelab Kit.Conclusions In HEV EIA kit the coating antigen should contain the ORF2 protein N-terminal antigen site of HEV genotype 4 which may increase the sensitivity of detection for HEV infection.
