摘要
目的 证实人淋巴细胞表面有钙调素结合位点存在。方法 用 10nm胶体金标记重组人钙调素 ,与人淋巴细胞反应后 ,用透射电镜观察。结果 实验组 (金标记钙调素 )淋巴细胞表面可见散在金颗粒 ;对照组 (用金标记BSA代替金标记钙调素或淋巴细胞预先用未标记钙调素封闭后再与金标记钙调素反应 )细胞表面未见金颗粒。结论 用免疫金标记透射电镜观察证实 。
Objective To confirm the existence of calmodulin binding-site on lymphocyte surface.Methods Fresh human peripheral lymphocyte was incubated with recombinant human calmodulin (rhCaM) labeled 10nm colloidal-gold, then observed using transmission electron microscope.Results There was gold particles dispersed on lymphocyte surface after incubated with rhCaM-labeled colloidal-gold, while no gold particle was found in the two control experiments.Conclusions This immuno-glod transmission electron observation directly verified the existing of calmodulin binding-site (CaM-receptor?) on lymphocyte surface.
