摘要
目的 :研究三七总皂甙 (PNS)对腹主动脉缩窄大鼠心肌肥大的影响。方法 :用腹主动脉缩窄法建立压力超负荷性心肌肥大模型。实验分假手术对照组 (A组 ) ,腹主动脉缩窄组 (B组 ) ,5 0mgPNS (kg·d)ip组 ,10 0mgPNS (kg·d)ip组 ,15 0mgPNS (kg·d)ip组进行。 3周后检测全心重量 (HW)、左心室重量 (LVW )、HW 体重 (BW )、LVW BW即左心室重量指数(LVI)。左心室组织切片苏木精 伊红染色后测心肌细胞直径 (MD)。结果 :15 0mgPNS (kg·d)ip组的HW BW、LVI及MD显著低于B组 (P <0 0 1) ,10 0mgPNS (kg·d)ip组与B组比较MD显著降低 (P <0 0 5 ) ,HW BW、LVI也有下降 ,但无统计学意义(P >0 0 5 ) ,5 0mgPNS (kg·d)ip组与B组比较HW BW、LVI及MD均无统计学意义 (P >0 0 5 )。结论 :一定剂量范围的PNS能抑制腹主动脉缩窄大鼠心肌肥大的形成 ,浓度越高 。
Objective:To study the effect of total saponins of panax notoginseng(PNS)on cardiac hypertrophy of abdominal aorta-constricted rats.Methods:The cardiac hypertrophy of rats due to pressure overload was induced by constricting of abdominal aorta.There were five groups in our experiments,i.e.sham operated control group(Group A),aorta-constricted group(Group B),Groups of 50mg PNS/(kg·d)ip,100mg PNS/(kg·d)ip,150mg PNS/(kg·d)ip.Three weeks later,the heart-weight(HW),left ventricular weight(LVW),the ratio of HW/BW and LVW/BW(LVI)were measured.Then the cardiomyocyte diameter(MD)was measured,after stained by Hematoxylin-eosin.Results: HW/BW,LVI and MD of Group 150mg PNS/(kg·d)ip were significantly lower than those of Group B(P<0.01).The MD of Group 100mg PNS/(kg·d)ip was also significantly lower than that of Group B(P<0.05).The HW/BW,LVI of Group 100mg PNS/(kg·d)ip were not obviously influenced(P>0.05).There was no significant difference in Group 50mg PNS/(kg·d)of HW/BW,LVI and MD(P>0.05),compared with Group B.Conclusion:PNS can prevent cardiac hypertrophy induced by pressure overload in rats at the concentration of 100,150mg PNS/(kg·d).
