大鼠自体血管搭桥后血管桥VEGF mRNA表达及意义

摘要 目的 :探讨搭桥后血管桥VEGFmRNA表达情况 ,为临床预防内膜增生及血管桥的选择提供理论依据 .方法 :建立大鼠自体血管移植模型 ,每组 80只 ,分成静脉组 (V组 ) ,游离动脉组 (A组 ) ,非游离动脉组 (nA组 ) ,分别于不同时间切取搭桥血管 .应用免疫组织化学和原位杂交方法进行指标检测 .结果 :V组织内膜增生比A组明显 ,V组VEGFmRNA及蛋白信号明显比A组弱 (2 8%vs 5 6 % ,P <0 .0 1 ) .结论 :自体血管移植后VEFGFmRNA表达异常 ; AIM: The mechanism of intimal hyperplasia (IH) and the reason that the prolonged patency of the artery bypass is superior to the venous bypass remain unclear. To elucidate the mechanism of intimal hyperplasia after bypass procedures, we designed this study to investigate the expression of VEGF mRNA in the bypss. METHODS: Rats with autogenous vascular replacement were divided into venous group (V group), free arterial group (A group), and non free arterial group, with 80 in each group. The vascular grafts were harvested at different time. The specimens were detected by immunohistochemical technique and in situ hybridzation (ISH) to measure the expression of VEGF mRNA and protein in the neointimal. RESULTS : IH in V group was more obvious than that in A group (28% vs 56%, P <0.01). The expression of VEGF mRNA and protein in V group was lower than that in A group. CONCLUSION: The neointimal formation had significant correlation with the abnormal expression of VEGF mRNA and protein after autogenous vascular replacement. The increased expression of VEGF mRNA will contribute to the long patency of bypass.
