摘要
目的 :探讨Spurr树脂包埋剂是否适合制备血样电镜样品。 方法 :多次离心技术制备血样 ,并对原始的Spurr配方进行改良。改变增塑剂乙二醇二甘油醚的量 ,调节包埋块的硬度 ,使其适合制备体液样品的电镜超薄切片。结果 :超薄切片上红、白细胞排列紧密 ,每个视野可见多个细胞。与常规的Epon树脂包埋剂相比 ,明显增加了可供观察的信息量。细胞的结构清晰 ,未见明显的聚合损伤以及人工假象等。结论
Objective: To explore whether Spurr resin embedding meddium is suitable for making the electron microscopic specimen of blood. Methods: Multi centrifugation was used for preparing blood specimen, and the original Spurr prescription was modified. The solidity of embedding specimen was adjusted, in order to make it suitable for preparing ultrathin sections of blood, by changing the volume of DER 736. Results: On the ultrathin section, the red and white blood cells were close together, and many cells could be seen in each field of vision. Compared with the ordinary Epon resin embedding medium, the observing information was increased obviously in Spurr medium. In addition, the cellular structure was clear and no embedding damage and artificial psudophenomenon were found in Spurr embedded specimen. Conclusion: Spurr resin is a good embedding medium for making the electron microscopic specimen of blood.
