摘要
目的 探求在大鼠肝硬化形成的早期 ,红细胞电泳率和红细胞膜胆固醇含量的变化情况。方法 将Wistar大鼠随机分成对照组和肝硬化组 ,其中肝硬化组又分成 2、5、7、10周组 ,每组 10只。肝组织行H E和天狼猩红染色 ,下腔静脉取血检测红细胞电泳率和红细胞膜胆固醇的含量。结果 在本实验中 ,肝硬化 7w鼠的肝组织中有大量的胶原纤维增生并开始形成假小叶。对照组和 2w肝硬化组间的红细胞电泳率的差异不明显 (P >0 .0 5 ) ,而自给药的第 5周起出现明显的增加 ;肝硬化 2w鼠红细胞膜胆固醇含量明显升高 ,至第 5周开始降至正常水平以下。结论 本大鼠肝硬化模型的红细胞电泳率和红细胞膜胆固醇含量在第 5周后出现明显的异常 ,比形态学上出现假小叶等典型的肝硬化表现约提前了 2周时间 ,为临床肝硬化的早期诊断治疗提供实验依据。
Objective To study the changes of erythrocyte electrophoresis rate and erythrocyte membrane cholesterol concentration at early stages in liver cirrhosis rat.Methods Rats were divided into control and hepatocirrhosis groups,and the latter was also divide dinto 2w,5w,7w and 10w groups,10 rats in each group.Using strains of H-E and Sirius Red,the degree of liver cirrhosis was evaluated.Blood came from the inferior vena cava and erythrocyte electrophoresis rate and erythrocyte membrane cholesterol concentration were examined.Results A large amount of hyperplastic collagenous fiber was presented in liver tissue and the frame of the pseudolobuli was formed in 7w-hepatocirrhosis rats.In erythrocyte electrophoresis rate,no significant difference was found in control and 2w-hepatocirrhosis groups(P>0.05),and significant increase was appeared in 5w-,7w-,and 10w-hepatocirrhosis groups.The erythrocyte membrane cholesterol concentration in 2w-hepatocirrhosis group was significant higher than that in control group (P<0.05),and significanty lower than in 5w-,7w-,and 10w-hepatocirrhosis groups.Conclusion In the hepatocirrhosis models,the abnormity of the erythrocyte electrophoresis rate and the erythrocyte membrane cholesterol concentration of rats was 2 weeks earlier than that of the form of pseudolobuli in morphology.The experimental evidences were presented for early diagnosis and treatment to liver cirrhosis.
