中西药消除耐药质粒pR_(ST98)的研究

目的 用SouthernBlot杂交法检测中、西药在体外消除肠道杆菌携带的耐药质粒pRST98的结果。方法 用氧氟沙星和中药双黄连对携带pRST98的伤寒杆菌、大肠杆菌和鼠伤寒杆菌进行亚抑菌浓度的质粒消除试验 ;用地高辛标记的氨苄青霉素和四环素耐药基因探针进行SouthernBlot。结果 SouthernBlot杂交结果显示两种药物作用后pRST98上的耐药基因并未丢失 ,但同一质粒耐药基因在不同宿主菌中表达不同 ,导致最小抑菌浓度 (MIC)变化有明显差异。结论 氧氟沙星和双黄连能降低pRST98宿主菌的耐药性。

Objective Using Southern Blot to explore the possibility of cruing the pR ST98 in different enteric bacilli using Oflocaxin and Chinese traditional medicine Shuang huang-lian.Methods Sub-inhibitory concentration of the drugs was selected to crue the plasmid carried by pR ST98 . Southern Blot of the Amp and Tc probe were used to identify the drug resistant genes on pR ST98 .Results Southern Blot results showed that the drug resistant genes were still presented on pR ST98 , Oflocaxin and Shuang huang-lian could not eliminate pR ST98 in vitro. But the kinds of resistant markers coded by the plasmid were reduced and the level of drug resistance (MIC) had declined remarkably in some of the bacteria strains.Conclusion These two drugs could reduce the drug resistant level of pR ST98 hosted bacteria.
