挤压伤后脊髓神经元NT-3和NT-4的表达变化 被引量:5

摘要 为了研究脊髓挤压伤后 ,脊髓神经元 NT-3和 NT-4的早期变化 ,本研究采用免疫组织化学 ABC法和阳性神经元计数 ,结果在大鼠脊髓证明了 NT-3免疫阳性反应产物主要在胞核着色 ,NT-4则胞浆及胞核均着色。腹角的 NT-3阳性神经元数在脊髓挤压伤后 7d组和 2 1d组较正常组和 2 4h组明显增高 ( P<0 .0 1) ;背角的 NT-3阳性神经元仅 2 1d组较正常组有显著增加 ( P<0 .0 1)。腹角的 NT-4阳性神经元数在损伤后 2 4h组、7d组和 2 1d组均较正常组有显著增加 ( P<0 .0 5 ) ,且随时间的延长呈增加趋势 ( P<0 .0 5 ) ;背角的 NT-4阳性神经元数 7d组和 2 1d组较正常组和 2 4h组显著增加 ( P<0 .0 1) ,随时间延长仍有增加趋势 ( P<0 .0 1)。结论 :在脊髓挤压伤后的早期腹、背角 NT-3和 NT-4阳性神经元数均有增多 ,提示内源性 NT-3和 To explore the expression changes of NT 3 and NT 4 in the spinal cord neurons in early stage following crushed spinal cord injury (cSCI), immunohistochemical ABC staining and the positive neurons counting methods were used on the 20 μm transections of rat. The resuet showed that NT 3 positive products were mainly distributed in nucleus and NT 4 positive products were distributed in nucleus and cytoplasm. The 7d and 21d groups were significantly higher than the normal and 24 h groups on the NT 3 positive neurons counting in the ventral horn following cSCI( P<0.01), only 21 d group was significantly higher than the normal group in the dorsal horn (P>0.01). But 24 h, 7 d and 21 d groups were significantly higher than normal group on the NT 4 positive neurons counting in the ventral horn following cSCI (P>0.01), and the 7 d and 21 d groups were significantly higher than the normal and 24 h groups in the dorsal horn(P<0.01). There was a trend of increase with time following cSCI (P>0.01). The results indicate that the number of NT 3 and NT 4 positive neurons is increased in the spinal cord ventral and dorsal horns following cSCI, and the increased endogenous NT 3 and NT 4 may play an role in early repairment of nerve injury following cSCI.
