摘要
目的 :研究骨髓间质干细胞 (mesenchymalstemcells ,MSCs)的免疫调节作用。方法 :从胚胎股骨中分离培养间质干细胞 ,经克隆化分选出形态均一的细胞 ,用流式细胞术检测其表面标志以鉴定其纯度。将分离培养的骨髓间质干细胞接种于 96孔板 ,待其长满孔面积的 80 %~ 90 %时用直线加速器以 30GY剂量照射 ,然后与分离的外周血淋巴细胞共培养 3d ,以单独培养的淋巴细胞为对照 ,加入ConA刺激 6 8h ,MTT加入后继续温育 4h ,用酶标仪检测以观察MSCs对外周血淋巴细胞增殖反应的影响。结果 :MSCs对外周血淋巴细胞的转化有明显抑制作用 (抑制率为 2 8% )。结论 :MSCs具有抑制体外淋巴细胞增殖的作用。
Objective: To investigate the influence of mesenchymal stem cells (MSCs) on the regulation of immune function. Methods: MSCs were isolated and cultured from embryonic bone marrow. Selected surface antigen of MSCs were detected by flow cytometry and their morphologic characteristics were determined by microscopy. MSCs were plated in 96-well dishes. When they covered the 80%~90% place of well, they were irradiated with 30GY by the linear accelerator, and co-cultured for 3 days with peripheral blood lymphocytes (PBLs). PBL non-co-cultured with MSCs served as control. When the PBLs stimulated by ConA for 68 hours, MTT(5 mg/ml) was added to each well and continued to culture for 4 hours and then the PBL proliferation was measured by ELISA READER. Results: The MSCs significantly inhibited peripheral blood lymphocyte proliferation stimulated by ConA. The rate of inhibition was 28%. Conclusion: MSCs can suppress lymphocyte proliferation in vitro.
