猪逆转录病毒与生物人工肝及细胞移植

摘要 生物人工肝是近年国内外治疗重型肝炎及肝脏功能衰竭的研究热点,猪肝细胞因其来源广泛、功能稳定、解剖及生理与人相近、不易患病等优点成为目前生物人工肝及细胞移植使用较多的生物材料之一,但自报道猪内源性逆转录病毒(porcineendogenousretrovirus熏PERV)感染体外培养人细胞以来,跨种族间感染越来越引起人们的重视。本文拟就该病毒在生物人工肝及细胞移植中的研究进展综述如下。 Bioartificial liver is a new therapy for the treatment of hepatic failure. Porcine hepatocytes, including transgenic pig, have become a source of cells widely used in bioartificial liver and cell transplantation because of their anatomical and physiological similarities with humans. Since vitro infection of various human cell lines with PERV during cocultivation with porcine cell lines or primary cell cultures has been demonstrated, concerns have been raised about the satety of this potential therapy, especially the possibility of cross species transmission of porcine retroviruses. The present article will summarize the latest progress on it.
