摘要
目的 :探讨中性粒细胞在光气中毒肺损伤中的作用 .方法 :80只雄性昆明小鼠随机分为 4组 .环磷酰胺组与环磷酰胺 /光气组腹腔注射环磷酰胺 ,对照组与光气组ip生理盐水 ;3d后染毒 .每组 1 0只观察死亡率 ,另 1 0只 4h后断头处死 ,采血取肝肺 ,肺测湿干比和髓过氧化物酶 (MPO)活性 ,血清测丙二醛 (MDA) .结果 :对照组与光气组相比 ,肺MPO活力差异非常显著 (P <0 .0 0 1 ) .环磷酰胺 /光气组与光气组相比 ,死亡率差异非常显著 (P <0 .0 1 ) ,湿干比和肺MPO活性均差异显著 (P <0 .0 5 ) ,血清MDA虽然降低但不显著 .结论 :光气中毒后肺MPO活性显著增加 ,提示肺部有明显的PMN聚集 。
AIM: To study the effects of polymorphonuclear (PMN) leukocyte on mouse lung injury after phosgene exposure. METHODS: Eighty mice were randomly divided into four groups. Cyclophosphamide (CPA) group and phosgene plus CPA group were injected (intra peritoneal, ip) with CPA, and control group and phosgene exposure group were injected with 9 g/L NaCl 3 days before exposure. Ten mice from each group were killed to observe mortality and 4 h later, the rest 10 mice from each group were killed and their lungs were used to measure wet to dry weight ratio myeloperoxidase vigor (MPO), and fluorometry was employed to measure the blood serum malondialdehyde (MDA). RESULTS: The difference of MPO vigor between the control group and the other two groups treated by phosgene was very significant ( P <0.001), and the mortality of phosgene plus CPA group was significantly lower than that of phosgene group( P <0.01). The lung wet to dry weight ratio and the MPO vigor of phosgene plus CPA group were also lower than those of the phosgene exposure group( P <0.05). The blood serum MDA of phosgene plus CPA group was lower than that of the phosgene exposure group, but no significant difference was found. CONCLUSION: The MPO vigor, following phosgene exposure, increases greatly in lungs, indicating an obvious PMN aggregation. Depletion of PMN can relieve acute lung injury induced by phosgene.
