Isocorydine(Isoc) is an alkaloid isolated from the Chinese herb Dactylicapnos Scandens. Its main pharmacological effect is as a nonspecific vasodilator.
Ten rhesus monkeys wighing 7.0+2.0 kg(mean± SD) were bled 152.5± 57.4ml (27.2% of the total blood volume) from the carotid artery to cause hemorrhagic shock. Five monkeys were injected with 5% Isoc (2.5rnl/kg)iv and the other five with the same volume of physiological saline. We observed the changes of the shock for 20 minutes, and then transfused blood for restoration. The femoral arterial pressure, systolic (FAPs) and diastolic (FAPd), auricula arterioles pulse (AAP), femoral arterial blood flow (FAQ) and heart rate (HR) were recorded with an RM-6000 8 channel polygraph. The data were statistically calculated with two-ways analysis of variance and t test.
The experimental results showed that in the hemorrhagic shock monkeys, Isoc could improve arterioles function and cardiac muscle contractility and increase cardiac output, FAP and FAQ, but decrease HR. The effects of Isoc as compared with physiological saline were significant (P<0.05). Furthermore, the transfusion effect was better in the Isoc group than in the saline group. Therefore, we consider tLat Isoc is valuable for anti-hemorrhagic shock.
