摘要
运用正交实验设计探讨银杏GinkgobilobaL.外种皮中黄酮类化合物的提取工艺。由极差分析和方差分析得知,在本实验所考察的影响因素中,提取次数对提取的影响最大,温度对提取的影响次之,乙醇浓度对提取的影响最小。其中提取次数和提取温度对提取的影响达到显著水平,乙醇浓度对提取的影响没有达到显著水平。实验结果表明,确定银杏外种皮黄酮类化合物的提取条件为:70℃,7倍体积的60%乙醇,回流提取4次,每次1h。在上述条件下,银杏外种皮黄酮类化合物的提取率达到84%以上。分光光度法测定总黄酮的平均回收率为97 3%,5次平行的相对标准偏差CV为0 73%。
The optimum extracting conditions of flavonoids in the testa of Ginkgo biloba L. were investigated by using the factorial and the orthogonal design.From the variance analysis,it is known that extraction times,temperture and concentration had different effects on extraction.Among the three factors,extraction times and temperature had an obvious effect on extraction.The effect of concentration EtOH on extraction didn't reach an obvious level.The results showed that at the conditions of 70℃,7∶1 of 60% ethanol to the testa of Ginkgo biloba L.weight ratio,extraction for 4 times and 1h each time,the average extraction rate was as high as 84%.The average recovery of total flavonoids determined by spectrophotometry was 97.3 % and the coefficient of variation was 0.73%.
