摘要 目的 了解全身炎症反应综合征 (SIRS)早期各时段血浆肿瘤坏死因子 (TNF α)和白介素 6 (IL 6 )的变化及其在脾脏中的mRNA表达 ,探讨SIRS失控演变过程中前炎症细胞因子发挥的作用。方法 制作SIRS实验动物模型 ,按时相不同分组 ,分别于 2h、4h、6h和 8h处死动物 ,用ELISA法测定血浆TNF α和IL 6含量 ,RT PCR技术检测大鼠脾脏组织中TNF α、IL 6mRNA的表达。结果  2h后血浆TNF α含量即开始明显升高 ,4h时到达峰值 ,其后 6h、8h仍保持高水平 ;IL 6含量的变化与TNF α则明显不同 ,呈逐步上升趋势。脾脏组织中TNF α、IL 6mRNA的表达与以上结果相一致。结论 SIRS大鼠中 ,TNF α是早期释放的炎症细胞因子且在血浆中全期呈高水平 ,它可能在SIRS发生和发展中起非常重要的作用 ,而IL 6峰值靠后 ,在SIRS早期IL 6不是引起损伤的主要炎症因子。 Objective To inquire the changes of TNF-α and IL-6 in different time at early stage in SIRS rat serum,and the expression of mRNA in spleen,and discuss the role of pro-inflammatory cytokines in the progress of SIRS.Methods The SIRS rat models were made and divided into the groups according to time phase.The rats were killed at 2h,4h,6h and 8h,the consistency of TNF-α and IL-6 in serum were checked by enzyme-linked immunoadsordent assay(ELISA).The expression of TNF-α,IL-6mRNA in spleen were analysed by RT-PCR.Results The consistency of TNF-α in serum was increased obviously at once in 2h,and reached the peak in 4h,keeping high level in 6h and 8h,there were different changes between IL-6 and TNF-α,it displayed the tendency elevated gradually.The expression of TNF-α,IL-6 mRNA in spleen were conuplied with the results.Conclusion Discharging early and keeping at high level at early stage of SIRS rat,TNF-α was raised to get up very important action in the SIRS occurrence and development.The IL-6 peaking lag confirmed IL-6 is not the main inflammatory cytokine causing the injury in early stage of SIRS.
