摘要
目的 :制备抗BACP1多克隆抗体并鉴定其特异性 ,用于BACP1蛋白定位及功能研究 .方法 :大肠杆菌表达GST BACP1融合蛋白 ,采用纯化蛋白免疫新西兰大白兔制备抗血清 .采用WesternBlot和间接免疫荧光染色等方法鉴定抗体的特异性 .结果 :抗BACP1抗体用于WesternBlot和间接免疫荧光染色均可特异性识别BACP1蛋白 ,具有较高的特异性 .结论 :所制备的多克隆抗体具有较高特异性 。
AIM: To prepare the anti BACP1 polyclonal antibody and identify its specificity. METHODS: GST BACP1 fusion protein was expressed and purified from E. coli . Zelanian rabbits were immunized using the fusion protein and the antiserum was prepared. Western Blot and immunohistochemical experiments have been performed to identify the antibody’s specificity. RESULTS: This polyclonal antibody can recognize and bind BACP1 specifically in vitro and in vivo . CONCLUSION: This anti BACP1 antibody has high specificity and affinity with BACP1 and would be a very useful tool for study of the BACP1’s localization and function further.
