高温合并化疗对耐药性VX-2细胞多药耐药基因的影响

摘要 目的 :观察热化疗对多药耐药性VX 2细胞mdr基因的影响 .方法 :采用单纯化疗 (阿霉素 32 μg/mL ,1h) ,单纯热疗 (4 2 .5℃ ,1h)及热疗联合化疗 (阿霉素 32 μg/mL +42 .5℃ ,1h) ,分别作用于VX 2及VX 2mdr1细胞 .观察不同处理组细胞的存活率、mdr1mRNA及P gp的表达情况 .结果 :热化疗组细胞的存活率明显低于单纯加热组及单纯化疗组 ,同时热化疗组细胞mdr1mRNA的表达明显减低或消失 ,P gp表达明显减低 .结论 :热化疗可以使多药耐药细胞mdr1mR NA表达降低或消失 ,使P gp表达降低 ,从而逆转了肿瘤细胞的多药耐药性 。 AIM: To observe the effect of thermochemotherapy on VX 2 mdr 1 cells MDR gene. METHODS: VX 2 cells received hyperthermia (42.5℃ for 1 h), chemotherapy (ADM 32 μg/mL) and hyperthermia plus chemotherapy (42.5℃ for 1 h + ADM 32 μg/mL) respectively. The survival rate of cells and the expression of mdr 1 mRNA and P gp after thermochemotherapy were determined. RESULTS: The survival rate of the cells receiving hyperthermia plus chemotherapy was significantly lower than that of the cells receiving hyperthermia and chemotherapy respectively. After hyperthermia puls ADM, a lower (or lost) expression of mdr1 mRNA and lower expression of P gp of VX 2 mdr 1 cells were found. CONCLUSION: Thermochemotherapy can decline tumor cell's expression of mdr1 mRNA and P gp, reverse tumor cell's MDR, enhance the sensitivity of tumor cells to drugs.
