β-七叶皂甙钠对大鼠短暂性局灶性脑缺血损伤的保护作用 被引量:5

摘要 目的 :探讨β 七叶皂甙钠对大鼠局灶性脑缺血损伤的保护作用 .方法 :4 0只雄性SD大鼠 ,随机分为 4组 ,即对照组 ,β Ae 5 ,β Ae 1 0和 β Ae 2 0保护组 ,保护组大鼠脑缺血前30min腹腔注射 β 七叶皂甙钠注射液 ,剂量分别为 5 ,1 0 ,2 0mg/kg ,对照组腹腔注射等量的生理盐水 .动物模型采用右侧颈内动脉线栓法致大脑中动脉阻闭 1 2 0min ,术后观察 2 4h神经行为学变化并评分 ,2 4h处死动物 ,取大脑行TTC染色以测量脑梗死容积 .结果 :术后动物均表现一定神经功能障碍 ,2 4h内对照组神经功能障碍逐渐加重 ,β 七叶皂甙钠保护则呈减轻趋势 .2 4h神经功能评分 ,β 七叶皂甙钠保护组分别为 (1 .2± 0 .9) ,(1 .1± 0 .7)和 (1 .2± 0 .8) ,明显低于对照组 (2 .1± 0 .6 ) (P <0 .0 5 ) .脑梗死容积 β 七叶皂甙钠组分别为 (87.9± 2 1 .3)mm3 ,(6 8.2± 1 5 .2 )mm3 和 (6 3.8± 1 8.3)mm3 ,均明显小于对照组 (1 95 .8± 31 .3)mm3 (P <0 .0 5 ) .结论 :β AIM: To study the protective role of β aescinate against focal cerebral ischemia in rats. METHODS: Forty male SD rats (body weight 280-320 g) were randomly divided into 4 groups ( n =10 each): Control group, Group β Ae 5, Groupβ Ae 10 and Groupβ Ae 20. The rats in the Control group were injected 2 mL saline, while the rats in the other three groups were injected 5 mg/kg, 10 mg/kg and 20 mg/kg β Aescinate 30 min before middle cerebral artery occlusion (MCAO) with 3 0 nylon monofilament, respectively. MCAO lasted 120 minutes. The neurological outcome of each group was evaluated at 24 h after operation. The infarct volume of each group was then assessed by TTC staining. RESULTS: The neurologic deficit scores (NDS) of Group β Ae 5 (1.2±0.9), Group β Ae 10 (1.1± 0.7), and Group β Ae 20 (1.2±0.8) at 24 h were lower than that of the Control group (2.1±0.6), P <0.05. The infarct volume of Control group (195.8±31.3) mm 3 was significantly larger than those of the other three groups[β Ae 5:(87.9±21.3) mm 3, β Ae 10: (68.2±15.2) mm 3,β Ae 20: (63.8±18.3) mm 3] at 24 h after operation. There was no difference among the β Ae treated groups in NDS and infarct volume. CONCLUSION: β Aescinate injection significantly reduces neurologic injury induced by transient MCAO without dose response effect.
