摘要
目的 :检测葡聚糖结合蛋白A[Glucanbindingprotein·A(GBPA) ]-壳聚糖纳米颗粒载体系统经鼻黏膜免疫小鼠的效果。证明壳聚糖可作为经鼻用蛋白防龋疫苗的载体系统。方法 :用该载体系统经鼻免疫小鼠 ,间接ELISA方法检测血清中针对葡聚糖结合蛋白A的特异的IgG以及唾液中针对葡聚糖结合蛋白A的特异的IgA。与霍乱毒素 - (GbpA)、未免疫组、空白壳聚糖纳米颗粒组作比较。 结果 :产生的血清和唾液中针对GbpA的特异的IgG、IgA抗体滴度远大于未免疫组、空白壳聚糖纳米颗粒组 ,与霍乱毒素 - (GbpA)组抗体滴度相当。结论 :证明壳聚糖纳米颗粒系统可作为经鼻运输葡聚糖结合蛋白A的载体系统。
AIM:To investigate the specific antibody titres of GbpA-chitosan nanoparticles in serum and saliva as a novel nasal delivery system for anti-carious vaccine.METHODS:Determine serum anti-GbpA IgG and salivary anti-GbpA IgA titres,evaluate antibody level compare to positive control group and negative control groups.RESULTS:GbpA-chitosan nanoparticles as a novel nasal delivery system induced protective specific anti-GbpA antibodies and the antibodies level corresponds to CT-GbpA group's antibodies level.CONCLUSION:Chitosan nanoparticles have the ability to transport GbpA anti-carious vaccine by nasal route.
