大鼠垂体IL-2表达和组织定位及卵巢切除对其表达的影响

摘要 目的 :探讨在大鼠垂体前叶中是否有白细胞介素 2(IL 2 )表达 ,并明确其组织定位 ,以及卵巢切除 (OVX)对其表达的影响 .方法 :将大鼠分为正常对照组 (NOR)、卵巢切除组(OVX)、卵巢切除 +雌二醇组 (OVX +E)、卵巢切除 +油剂组(OVX +O)和假手术组 (SO) ,取垂体前叶标本进行免疫组化染色 ,并以图像分析方法分析垂体前叶细胞中IL 2免疫阳性物质含量的变化 .结果 :IL 2免疫反应阳性物质仅分布于前叶 ,在中叶和后叶则未见 .与正常对照组相比 ,OVX组和OVX +O组IL 2免疫阳性物质含量明显增多 ;而OVX +E组IL 2免疫阳性物质含量无明显变化 .结论 :我们首次报道IL 2在大鼠垂体的组织定位 ,机体雌激素水平下降可明显上调其表达 . AIM: To investigate whether there is IL 2 expression in the anterior pituitary and if any, to localize its position and to explore the effects of ovariectomy on its expression. METHODS: Immunohistochemical ABC method and image analysis were applied. RESULTS: IL 2 immunoreactive substances were only restricted in the anterior lobe, and not in the intermediate or posterior lobe. The immunostaining of IL 2 in the anterior pituitary was enhanced following ovariectomy, but it could be restored to the normal if supplemented with benzoate estradiol. CONCLUSION: To our knowledge, this is the first time to report that there are IL 2 immunoreactive substances localized in the anterior pituitary of rats, and decrease of estrogen level in the body can markedly upregulate the expression.
