摘要
目的 :研究氨基葡萄糖硫酸盐 (GS)对白血病细胞HL6 0的增殖和分化的影响 .方法 :采用台盼蓝活细胞计数法检测不同浓度的GS对HL6 0细胞增殖的影响 ;细胞化学染色法观察药物作用后细胞形态的变化 ;流式细胞仪检测药物对HL6 0细胞周期的影响及细胞表面分化抗原CD1 1b ,CD1 4 ,CD1 3和CD33表达的影响 .结果 :GS能明显抑制HL6 0的增殖 ;经 5mmol/LGS处理 5d后 ,HL6 0细胞体积缩小 ,核浆比例降低 ,核仁减少甚至消失 ,核染色质趋向致密 ,核形态扭曲折叠或分叶 ;5mmol/LGS处理 4 8h后 ,G1期细胞由 37.8%增加至 4 7.9% ,S期由 4 6 .8%减低至 4 0 .6 % ,G2期峰轻度减低 ,未发现亚二倍体峰 .经 1mmol/L ,5mmol/LGS分别处理 1 2 0h后 ,CD1 1b和CD1 4的表达分别由 2 .3% ,0 .5 %升高至 2 3.0 % ,1 1 .9%和 1 5 .8% ,4 .3% ,CD33及CD1 3的表达未见明显变化 .结论 :GS能抑制白血病细胞HL6 0的增殖 ,并诱导其向成熟单核。
AIM: To study the differentiation and inhibition of the proliferation of leukemia cell line HL60 induced by glucosamine sulphate (GS). METHODS: Using trypanblue, growth curves were drawn by vital cells number counting. Wright Giemsa staining was performed to observe the morphology changes of HL60 cells and flow cytometry was used to observe cell cycles and the expression of CD11b,CD14, CD13 and CD33, separately. RESULTS: The changes of the growth curve showed that the proliferation of HL60 cells was inhibited by GS. After treatment with 5 mmol/L GS for 48 hours, the percentage of G1 phase HL60 cells increased more than that of the untreated and the percentage of S phase cells decreased. No sub G1 was observed. After treated with 1 mmol/L or 5 mmol/L GS for 120 hours, the expression of CD11b and CD14 increased, but no significant changes of CD13 and CD33 expression were observed. CONCLUSION: GS can inhibit the proliferation of HL60 cells and induce the differentiation of HL60 cells along the granulocytic or monocytic lineage and the mechanism needs to be further studied.
