佛波酯对人早幼粒白血病细胞系细胞分化和凋亡的诱导作用 被引量:1

摘要 目的 :探讨佛波酯 (TPA)对人早幼粒白血病细胞系 (HL - 6 0 )分化和凋亡的影响。方法 :利用TPA体外培养HL - 6 0细胞 ,依据形态学和细胞化学的变化 ,DNA琼脂糖凝胶电泳和流式细胞仪等技术进行分析。结果 :7 2× 10 -6mol/L的TPA可使HL - 6 0细胞出现向单核 /巨噬细胞分化的形态特征 ,随培养时间延长 ,可见HL - 6 0细胞核固缩、染色体断裂、凋亡小体形成等细胞凋亡的形态学改变。TPA处理 2 4h可使酸性非特异酯酶 (α -NAE)的活性和四氮唑蓝 (NBT)还原能力显著增强 ,DNA琼脂糖凝胶电泳显示典型的“梯子”状带纹。TPA处理 0、2 4、4 8h凋亡细胞百分数分别为 (4 31± 1 2 3) %、(15 10± 1 31) %和 (2 3 2 0± 3 36 ) % ,在整个实验过程中细胞周期时相、Bcl-2蛋白含量无明显变化。结论 :TPA可诱导HL - 6 0细胞向单核 /巨噬样细胞分化与凋亡 ,且此凋亡过程可能与细胞周期时相、Bcl- 2蛋白无关。
