摘要
目的 探讨转化生长因子β(TGF-β)通路中 Smad4 蛋白与转化生长因子β 型受体 (TGF-β R )蛋白的变化及其在肝细胞癌中可能的作用机制。方法 用免疫组化 SABC法检测 4 5例石蜡包埋人肝细胞癌标本及36例癌旁组织中 Smad4 、TGF-β R 蛋白表达情况。结果 4 5例肝癌组织中 ,Sm ad4 、TGF-βR 阳性表达率分别为 5 1.5 %和 4 2 .2 %。肝细胞癌组织中 Smad4 、TGF- β R 阳性表达与癌旁组织比较均明显降低 (P <0 .0 5 ) ,且与包膜是否完整和有无癌栓有关 (P <0 .0 5 ) ,TGF- βR 与肝细胞癌组织分化程度有相关性 (P <0 .0 5 )。结论 肝细胞癌组织中 Smad4 、TGF- β R 阳性表达均降低 ,Smad4 及 TGF- βR 的丢失。
Objective To explore the relationship between Smad 4 and transforming grouth factor β receptor Ⅱ(TGF βRⅡ) in TGF β pathway and possible mechanisms by which they affect hepatocellular carcinoma Methods The expression of Smad 4 and TGF β RⅡ in proaffin embeded HCC tissues was detected by using antibodies against Smad 4 and TGF βRⅡ with SABC immunohistochemistry Results The postive rates of Smad 4 and TGF β RⅡ was 51 1% and 42 2% respectively There was a significant relationship between the expressin of Smad 4 or TGF β RⅡ and poorencapsulating or portal embolus of tumor (P<0 05) Conclusion The expressin of Smad 4 or TGF β R Ⅱ in HCC tissue are significantly decreased Smad 4 or TGF β RⅡ may help to evaluate prognosis of HCC
