摘要
目的 研究钙黏附分子 (E cadherin ,E cad)、α 连接蛋白 (α catenin ,α cat)和基质金属蛋白酶 (MatrixMetallopro teinase 2 ,MMP2 )在膀胱移行细胞癌中的表达及预后意义。方法 应用SP免疫组化染色法 ,检测 96例膀胱移行细胞癌和 10例正常膀胱黏膜E cad、α cat及MMP2的表达情况。结果 正常膀胱黏膜E cad、α cat呈正常表达 ,MMP2为阴性 ;96例膀胱移行细胞癌中E cad及α cat的异常表达率分别为 4 4 8% (43/96 )和 5 8 3% (5 6 /96 ) ,MMP2阳性表达为 5 3 1%(5 1/96 )。E cad和α cat的异常表达率、MMP2的阳性表达率与肿瘤的病理分级、临床分期及生存率显著相关。结论 E cad、α cat和MMP2在膀胱移行细胞癌的演变过程中起重要作用 ,可作为监测肿瘤恶性程度和判断预后的指标。
Objective To study the relation and clinical significance of E-cadherin (E-cad), α-catenin (α-cat) and matrix metalloproteinase 2 (MMP2) expression in bladder transitional cell carcinoma. Methods Immunohistochemical staining (streptavidin-peroxidase method) was used to detect E-cad, α-cat and MMP2 expression in 96 cases of bladder transitional cell carcinoma and 10 normal bladder tissues. Results E-cad, α-cat were uniformly positive and MMP2 was negative in all of the 10 normal bladder tissues. The abnormal expression rates of E-cad and α-cat were 44.8%(43/96) and 58.3% (56/96) in 96 cases of bladder cancer, respectively. The positive rate of MMP2 was 53.1% (51/96) ( P <0.01). There was a positive relationship between the expression of E-cad and α-cat. The abnormal expression rates of E-cad, α-cat and positive expression rate of MMP2 showed a significant correlation with the pathologic grade, clincal stage, high recurrence and poor prognosis. Conclusions Expression of E-cad, α-cat and MMP2 plays an important role in the malignant progression of bladder transitional cell carcinoma and can be used as valuable molecular markers for the tumor progression, recurrence and prognosis.
