摘要
目的 :研究同型半胱氨酸 (HCY)、同型半胱氨酸诱导基因抗体 (HCY -2Ab)及叶酸对人胚心肌细胞HCY 2表达及对心肌细胞增殖的影响。方法 :采用免疫细胞化学、RT PCR法体外测定人胚心肌细胞内HCY 2表达 ;用细胞计数法和MTT法对心肌细胞增殖的变化定量分析。结果 :当HCY的浓度小于 1 .2 5mmol/L时 ,HCY 2的表达随着HCY浓度的增高而逐渐增强 ,HCY促进细胞的增殖 ;当HCY浓度超过 1 .2 5mmol/L时 ,则HCY 2的表达逐渐降低 ,HCY抑制细胞的增殖 ;加入HCY 2Ab或叶酸 ,HCY 2的表达减弱 ,同时抑制细胞的增殖。结论 :HCY 2高表达时HCY对心肌细胞的增殖起促进作用 ;HCY 2低表达时则HCY、HCY 2Ab或叶酸抑制心肌细胞的增殖。
Objective:To study the effects of homocysteine(HCY), homocysteine induced gene antibody(HCY-2Ab) and folic acid on HCY-2 expression and proliferation of human embryonic cardiac myocytes. Methods:Immunocytochemistry and RT-PCR were used to analyze the HCY-2 expression on cultured human embryo cardiac myocytes; the cell counting and MTT were used to analyze the proliferation of human embryo cardiac myocytes quantitatively. Results:When the concentration of HCY was less than 1.25 mmol/L, the expression of HCY-2 and the cell proliferation increased as the concentration of HCY increased. The cell proliferation and the expression of HCY-2 were inhibited when the HCY exceeded 1.25 mmol/L. When adding HCY-2Ab or folic acid, the expression of HCY-2 decreased and the cell proliferation were inhibited. Conclusion:Myocyte proliferation is stimulated by HCY when HCY-2 is highly expressed,and the proliferation is inhibited by HCY, HCY-2Ab and folic acid when HCY-2 is weakly expressed.
