摘要 目的 :观察氧化型低密度脂蛋白 (ox LDL)作用下丘脑垂体后条件培养液对内皮细胞丙二醛 (MDA)代谢和NOS表达的变化 ,探讨下丘脑血管中枢对动脉粥样硬化形成的影响。方法 :按ox LDL诱导下丘脑 垂体 (ox HP)条件培养液 ,培养正常及受损内皮细胞。收集上清液检测MDA含量 ,细胞用免疫细胞化学法检测NOS表达。结果 :( 1 )MDA含量 :实验组对正常内皮细胞下调 ;而对受损内皮细胞ox H和ox HP组的下调作用明显减弱。 ( 2 )NOS表达 :ox H和ox HP组对正常内皮细胞上调 ;对受损内皮细胞下调。结论 :ox LDL作用下丘脑垂体后条件培养液对正常内皮细胞起抗损伤作用和保护调节作用 ;对受损内皮细胞的抗损伤作用和保护机制减弱。 Objective:To observe the changes of MDA metabolism and NOS expression in endothelial cells treated by the hypothalamus-pituitary conditioned media and to investigate the influence of hypothalamic vessel center on pathogenesis of AS. Methods:The conditioned media were collected respectively according to hypothalamus(H), hypothalamus+pituitary(HP),hypothalamus induced by ox-LDL(ox-H), hypothalamus+pituitary induced by ox-LDL(ox-HP) and control. The normal and damaged EC were cultured separately with the conditioned media. Then the supernatants were collected respectively to detect the content of MDA and the cells were fixed for immunocytochemical staining of NOS expression. Results:(1)The conditioned media of all experimental groups showed down regulation in the contents of MDA in normal EC, while the down-regulation effect was obviously decreased in ox-H and ox-HP groups of the damaged endothelial cells. (2)The conditioned media of ox-H and ox-HP group showed mild up-regulation effect on the expression of normal EC,but had down-regulation effect on the expression of damaged EC. Conclusion:The hypothalamus-pituitary conditioned media induced by ox-LDL has protective effects on the normal EC and the protective effects on damaged EC is decreased.
