摘要
目的 建立一种肠道病毒的分子生物学检测方法 ,查明福建省肠道病毒的流行型别。方法 病毒分离株用RT-PCR法扩增目的基因片段 ,然后测序 ,将基因序列提交到NCBIGenBank上进行比对 (Blast) ,同时运用Bioedit软件进行同源性分析 ,并构建基因树。结果 发现FJ96 - 71病毒株与GenBank上的名为“untype”的毒株序列同源性较高 ,可能属于同一新型肠道病毒 ,FJ97- 6 3病毒与ECV1同属一血清型 ,可能为一新基因型。结论 运用分子生物学方法可以检测出传统方法所无法鉴定的血清型 ,还能提供其它方法所无法提供的病毒遗传学等方面的信息。
To establish molecular methods for enteroviruses typing and learn which types prevail in Fujian province.A VP1-specific fragment was amplified by RT-PCR with primer 011/012 and then sequenced.The serotype was determined by comparing the sequence of the VP1 amplicon to NCBI Genebank database.The identities was analysis and phylogenetic tree was constructed with Bioedit program.The results showed that FJ96-71 strain was hight identities with the 'untype' strains in genebank,suspiciously represented a new serotyped.FJ97-63 was homologous serotype with ECV1,suspiciously a new genotype.A molecular typing system based on such method can identify 'unknow' serotype strains by conventional methods and may provide additional biological information.
