ets-l反义寡核苷酸对胃癌细胞株SGC-7901体外侵袭力的影响

摘要 目的 :研究ets l反义寡核苷酸对胃癌细胞株SGC 790 1体外侵袭力的影响。方法 :应用ets l反义、正义寡核苷酸转染SGC 790 1细胞 ,并设PBS对照组。转染后应用逆转录聚合酶链式反应检测ets lmRNA的变化 ,应用克隆形成试验检测细胞增殖活性的变化 ,Transwell小室检测侵袭能力的变化。结果 :转染ets l反义寡核苷酸后 ,ets lmRNA表达降低 ,反义组形成克隆数目 (2 4 2± 4 8)较正义组、对照组减少 (47 6± 8 1,4 4 3± 7 6 ,P <0 0 1) ,侵过小室滤膜细胞数目减少 (97 1± 11 1,198 7± 18 3,2 0 5 8± 2 2 2 ,P <0 0 1)。结论 :ets l反义寡核苷酸能够抑制胃癌细胞株SGC 790 1的体外侵袭力。 Objective:To investigate the effect of ets 1 antisense oligoxydeonucleotide(ASODN)on invasive ability of gastric cancer cell line SGC 7901 in vitroMethods:Cultured cells were divided into three groups:control group,sense oligoxydeonucleotide(SODN)group and ASODN group.After transfection for 24h,cultured cells were harvested to detect the changes of ets lmRNA by the reverse transcription polymerase chain reaction method(RT PCR),growth inhibition was detected by colony forming unit assay,invasive ability was measured by the number of cells to penetrate polycarbonates coated with or without matrigel.Results:est 1 ASODN could underregulate the expression of ets 1mRNA,number of colony forming was significantly lower than the other two groups(24.2±4.8,47 6±8 1,44 3±7 6,P<0 01),number of cells that penetrated polycarbonates was also significantly lower(97 1±11 1,198 7±18 3,205 8±22 2,P<0 01)Conclusion:ets lASODN may inhibit invasive ability of gastric cancer cell in vitro.
