摘要
目的 :探讨米非司酮对早孕绒毛、蜕膜组织中胰岛素样生长因子_Ⅱ (IGF_Ⅱ )mRNA表达的影响及IGF Ⅱ与早期妊娠及流产的关系。方法 :采用逆转录聚合酶链式反应 (RT PCR) ,对 2 0例人工流产妇女 (对照组 ) ,2 0例服用米非司酮药物流产的早孕妇女 (药物组 )绒毛、蜕膜组织中IGF_ⅡmRNA的表达进行检测。苏木精 伊红染色 ,PAS染色分别观测其绒毛及蜕膜组织中组织形态学改变及其糖原储存量变化。结果 :药物组绒毛、蜕膜组织中IGF_ⅡmRNA的表达与对照组相比明显降低(P <0 0 5 ,P <0 0 1)。对照组绒毛、蜕膜组织中糖原含量丰富 ,药物组均较低 (P <0 0 5 ,P <0 .0 1)。结论 :米非司酮可能通过下调绒毛、蜕膜组织中IGF_ⅡmRNA的表达 ,影响胎盘功能 。
Objective:To investigate the effect of Mifepristone on the expression of insulin-like growth factor-Ⅱ(IGF-Ⅱ)mRNA in human villus and decidua,and the relationship of IGF-Ⅱ to early pregnancy and abortion. Methods: Villus and decidua samples were obtained from 20 medical abortion patients(medicine group)and 20 surgical abortion patients(control group). Quantitative reverse transcription-polymerase chain reaction(RT-PCR) assay was utilized to determine the level of mRNA expression of IGF-Ⅱ in two groups.The morphology and the glycogen content in all samples were investigated by histochemistry and image analysis technology. Results: The levels of IGF-ⅡmRNA in villus and decidua in medicine group were significantly decreased,compared with those in control group(P<0.05,P<0.01).The glycogen contents in villus and decidua in control group were abundant, but insufficient in medicine group(P<0.05, P<0.01). Conclusion:Mifepristone may influence placental function,and impede embryo growth by inhibiting the expression of IGF-Ⅱ gene in villus and decidua in early pregnancy.
